Comparative Analysis and Characteristic Verification of the Quality Standard of Milk Thistle Extract

There are some differences in the quality characteristics and standard values of milk thistle extract set by pharmacopoeias and standards in various countries and regions.
MethodsTo promote the standardization of quality standards for milk thistle extract, this paper summarized and compared the standard characteristics and related values settings in four different standards. At the same time, actual samples were tested for the characteristics that appeared more frequently in standard. We found that the ratio algorithm of silymarin set in the pharmacopoeia standards was not reasonable and could blur the quality of related products from the test results, especially without limiting the total amount of silymarin in the standard.
ResultsWe suggested setting standard values for all six silymarin characteristics, establishing more universal detection methods, and preparing relevant reference materials to match the detection methods. Based on the comparison of standards and test data, the characteristics of the ethanol content, ethyl acetate content, methanol insoluble substances, heavy metal content, and microbial characteristics should draw more attention and should be considered in the future for setting newly formulated standards.
ConclusionThis paper lays the data foundation for developing higher quality standards for milk thistle extract, which plays an important role in promoting the international trade of milk thistle extract.

Effect of Herbal Medicine on Insulin Signaling Pathways and Insulin Resistance in Metabolic Dysfunction-associated Fatty Liver Disease: A Review

EkanayakeIntroductionMetabolic dysfunction-associated fatty liver disease (MAFLD) is the most common chronic liver disease progressing towards a substantial global health concern. A multitude of therapeutic agents are being used to treat MAFLD. The high prevalence and adverse effects of drugs have increased the global popularity of herbal medicines (HMs) as effective therapeutic agents to treat MAFLD. Among the causes of pathogenesis, insulin resistance (IR) plays a crucial role in MAFLD. Hence, the amelioration of IR has emerged as a promising target for potential therapeutic approaches in MAFLD.
AimThis study aimed to explore in vitro and in vivo mechanisms that unveil recent advances in HMs that target the amelioration of IR in MAFLD.
Materials and MethodsElectronic databases, including PubMed and MEDLINE, were used to search literature for HMs in the management or treatment of NAFLD published up to March, 2023. The three primary search terms were “MAFLD” “NAFLD” and “HM”. MeSH of NAFLD, such as (Non-alcoholic fatty liver disease, Non-alcoholic Steatohepatitis) and HM (Medicinal plants, Plant extracts), and keywords were used to optimize the search strategy.
ResultsHMs act on a multitude of molecular pathways that increase insulin sensitization including upregulation of the secretion and activation of insulin receptor substrate (IRS) proteins, upregulation of phosphatidylinositol 3-kinase (PI3K)/AKT (also known as PKB or protein kinase B) signaling pathway, activation of the 5' AMP-activated protein kinase (AMPK) pathway and modulation of enzymes of glucose homeostasis, regeneration of the pancreatic β-cell mass, modulation of secretion of adipokines, enhancement of the uptake of glucose into tissues via increased expression of GLUT in tissues, and upregulation of FGF1/FGF1R signaling pathway.
ConclusionHMs possess a multitude of potential actions that reduce the IR and improve glucose homeostasis. Hence, HMs are a promising resource as effective medications for the treatment of MAFLD.

Research Progress in Natural Products against Liver Fibrosis
Authors: Siyu Zhao, Xin Chen, Rui Tan, Xiaoqing Wu and Hezhong JiangFibrosis of the liver is the most common pathological feature of many chronic liver diseases, progressing to cirrhosis and eventually liver cancer. Currently, only individual genes or proteins are targeted by synthetic drugs used to treat liver fibrosis, they have limited therapeutic effect, and are associated with many side effects. In contrast, natural products have shown great advantages and potential in the treatment of liver fibrosis due to their structural diversity, high multi-target activity and low toxicity and side effects. Therefore, this review concludes the pathological mechanism of liver fibrosis and the cellular and signaling pathways involved, provides new ideas for anti-liver fibrosis research and introduces common natural products and their anti-hepatic fibrosis mechanisms.

Targeting the Receptor Complexes by Structure-based Natural Drug Compounds in Seizures: A New Dimension in Drug Discovery and Design
A seizure is the brain's uncontrolled, abnormal electrical activity, which may result in altered states of consciousness, behavior, memory, or emotion. Seizures start biologically with the activation of susceptible brain neurons, which causes synchronized discharges of larger groups of connected neurons. A few potential causes of seizures include medications, genetics, electrolyte abnormalities, sleep state, infections, brain inflammation, and injuries. Medicinal plants are a rich source of various chemical molecules with distinct structures and biological activity. Most plants contain active components, including coumarin, glycosides, alkaloids, terpenoids, flavonoids, peptidoglycans, and other elements often associated with the effects of antiseizures. Isolating and identifying biologically active compounds and molecules from nature have resulted in the development of novel treatments, which in turn have contributed to the advancement of the health and pharmaceutical sectors throughout the history of humanity. In this review, we thoroughly summarize the information on the anti-seizure activities of medicinal plants and bioactive chemicals, focusing on molecular targets and cellular signaling pathways. All available research has contributed to medicinal plants as a reasonable option for seizure prevention and treatment, as well as drug development and manufacturing. To better comprehend the underlying molecular mechanisms, more research is required. If these mechanisms are discovered, it will be easier to identify new targets and create innovative anti-seizure therapeutic drugs to enhance patient survival and life quality. This work is expected to provide insights and ideas for the further research of Bioactive compounds from medicinal plants, their qualities, and the scientific basis for their improved clinical use.

Novel Approaches for the Extraction and Identification of Phytoconstituents from Herbs and Spices

Herbs and spices are now the primary ingredients in the global food and nutraceutical industry. Traditional methods for extraction of active metabolites of herbs and spices may lead to lowered extraction efficiency due to high solvent consumption and a longer extraction period. These issues can be resolved by the use of novel green and sustainable extraction techniques. The present paper aims to discuss innovative extraction and identification techniques for herbs and spices. Microwave-aided extraction (MAE), ultrasound-assisted extraction (UAE), supercritical fluid extraction (SFE), DNA barcoding, THz-S, e-nose, near-infrared (NIR) spectroscopy, hyperspectral imaging, Raman spectroscopy possess environment-friendly instrumentations, make lesser use of chemicals, and reduce the consumption of solvent. The use of cutting-edge technology in place of outdated ones can improve product quality and help the general public maintain high levels of health.

Systematic Detoxification of Copper-induced Toxicity by Methanolic Extracts of Anacyclus pyrethrum (L) in Zebrafish Model
IntroductionAnacyclus pyrethrum is used in Ayurveda to treat various ailments, ranging from toothache to female infertility. However, the molecular mechanisms of its medicinal properties are obscure and need exploratory studies.
MethodsWe aimed to investigate the molecular mechanisms associated with the medicinal properties of A. pyrethrum using zebrafish as an in vivo model. We have employed in silico, in vitro, and in vivo approaches to elucidate the anti-inflammatory, antioxidant, and cytoprotective functions of the methanolic extracts of A. pyrethrum. In silico analyses were carried out using Prediction of Activity Spectra for Substances (PASS), Molecular Inspiration, Swiss Absorption, Distribution, Metabolism, Excretion and Toxicity - ADME (SWISSADME), Absorption, Distribution, Metabolism, Excretion, and Toxicity - Structure-Activity Relationship(ADMETSar) and molecular docking tools to prove the anti-inflammatory effects of the bioactive components of A. pyrethrum.
ResultsOur results indicate that the bioactive components of A. pyrethrum have pharmaceutical properties with high intestinal absorption and greater bioavailability. Further, in vitro and in vivo results support the anti-inflammatory, antioxidant, and antitoxic effects of methanolic extract of A. pyrethrum via suppressing the levels of the inflammatory and Reactive oxygen species biomarkers, including Lactate Dehydrogenase, Myeloperoxidase, Nitric Oxide, and Lipid peroxidase respectively.
ConclusionThe results obtained from three different approaches in this study highlight the potential anti-inflammatory, antitoxic, and antioxidant effects of A. pyrethrum.
