This chapter describes the protocols to induce ocular hypertension in rabbits using silicone oil. This process consists of many steps and sub-steps, including preparation of the animal for anesthesia, preparation of materials, administration of anesthesia, preparation for silicone oil (SO) injection, intracameral injection of SO, and finally, postoperative care. Healthy animals free of ocular morphological abnormalities or disease states should be chosen for inclusion before beginning the protocol. Following these steps will allow for the simulation of ocular hypertension while also ensuring an aseptic environment.
