Abstract

Background and Objective: A range of strategies to eliminate cancer cells have been used, consisting of medical opera-tion, chemo- and radiotherapy. There is a significant issue in chemotherapy. The medications targeting delivery to object tissue cells are challenging because of the presence of numerous physical obstructions. Method: In the present study, we synthesized and characterized a novel and promising specific pHstimuli- responsive tumor-targeted DDS, polyethyleneimine (PEI) as a polymeric passive targeting agent, gluta-thione (GSH) as an active targeting agent as well as Capsaicin as a sensitizer and anticancer drug utilized as a theranostic agent. Results: The synthesized magnetic nanoparticles (MNPs) were fully characterized by FESEM, TEM, DLS, XRD and VSM. The morphology of MNPs has an acquired semi-spherical shape with specified diameters. Capsaicin is a successfully loaded MNP representing promising and admissi-ble biocompatibility as well as shows higher toxicity versus SK-N-MC cells in comparison with the control group. Furthermore, investigation proved the effectively internalized Drug Delivery System (DDS). In addition, the in vitro assay represented that the synthesized MNPs are nearly, fully pH stimu-liresponsive and release the Capsaicin at the controlled condition. Conclusion: MTT assay verified the higher toxici-ty of Capsaicin loaded into the MNPs in comparison with the control group. MRI in vitro investigation demonstrated that the prepared MNPs can be utilized as MRI imaging agents.
